Inhibition of Protein Synthesis in Mycobacterium smegmatis by a New Antibiotic Lividomycin

نویسنده

  • MICHIO TSUKAMURA
چکیده

Lividomycin inhibited incorporation of acetate-114C , glutamic acid-1-14C, leucine-1-14C, and glycine1-14C into protein of Mycobacterium smegmatis (strain Jucho). Incorporation of acetate-1-14C and glycine1-14C into nucleic acids was also inhibited, but incorporation of these compounds into lipids was not inhibited in the presence of lividomycin. Incorporation of uridine-2-14C into nucleic acids was significantly inhibited, but incorporation of thymidine2-14C into the nucleic acid fraction was not inhibited

MICROBIAL TRANSFORMATION OF CHOLESTEROL BY MYCOBACTERIUM SMEGMATIS

Mycobacterium smegmatis PTCC 1307 (CIP 73.26) was used as a microbial agent to produce androsta-1,4-diene-3,17-dione (ADD) and androst-4-ene-3,17-dione, two useful precursors in the synthesis of steroid drugs. The side chain of cholesterol, as the substrate, was selectively cleaved in the presence of five enzyme inhibitors. An intermediate structure with intact side chain, cholest-4-ene-3-one, ...

Reconstitution of Protein Translation of Mycobacterium Reveals Functional Conservation and Divergence with the Gram-Negative Bacterium Escherichia coli

Protein translation is essential for all bacteria pathogens. It has also been a major focus of structural and functional studies and an important target of antibiotics. Here we report our attempts to biochemically reconstitute mycobacterial protein translation in vitro from purified components. This mycobacterial translation system consists of individually purified recombinant translation facto...

The cytochrome bd-type quinol oxidase is important for survival of Mycobacterium smegmatis under peroxide and antibiotic-induced stress

Targeting respiration and ATP synthesis has received strong interest as a new strategy for combatting drug-resistant Mycobacterium tuberculosis. Mycobacteria employ a respiratory chain terminating with two branches. One of the branches includes a cytochrome bc1 complex and an aa3-type cytochrome c oxidase while the other branch terminates with a cytochrome bd-type quinol oxidase. In this commun...

Evaluation of DNA primase DnaG as a potential target for antibiotics.

Mycobacteria contain genes for several DNA-dependent RNA primases, including dnaG, which encodes an essential replication enzyme that has been proposed as a target for antituberculosis compounds. An in silico analysis revealed that mycobacteria also possess archaeo-eukaryotic superfamily primases (AEPs) of unknown function. Using a homologous recombination system, we obtained direct evidence th...

Mechanism of action of rifampin on Mycobacterium smegmatis.

Deoxyribonucleic acid (DNA)-dependent ribonucleic acid (RNA) polymerase (EC 2.7.7.6) isolated from a rifampin-sensitive strain of Mycobacterium smegmatis was 90% inhibited by 1 mug of rifampin per ml; enzyme from a rifampin-resistant mutant was not affected by this concentration of antibiotic. Inhibition of phenylalanine-1-(14)C incorporation by rifampin in growing cultures was complete about 6...
